Carboxyfluorescein succinimidyl ester (CFSE) is currently the most widely used green fluorescent probe for studying cell proliferation by in vitro or in vivo labeling of cells. CFSE spontaneously couples to both intracellular and cell-surface proteins by reaction with lysine side chains. When cell divide, CFSE labeling is distributed equally between the daughter cells, which are therefore half as fluorescent as the parents. Distributed CFSE labeling is readily detected by a flow cytometer, fluorescent reader or fluorescence microscope. The fluorescent CFSE has excitation/emission maxima ~492/517 nm.
